Discovered 90 years ago, the molecule Nicotinamide Adenine Dinucleotide (NAD, or rather\u00a0<strong>NAD+<\/strong>), also known as\u00a0<strong>coenzyme 1<\/strong>, is present in all cells and plays a central role in\u00a0<strong>metabolism<\/strong>, that is, the biochemical processes that lead to energy production. NAD+ influences several different cell functions, playing an important part in\u00a0<strong>DNA repair<\/strong>,\u00a0<strong>immune cell function<\/strong>, and\u00a0<strong>aging<\/strong>.<\/p>\n<h3><span style=\"color: #333333; font-size: 30px;\">Age goes up, NAD goes down<\/span><\/h3>\n<\/article>\n<\/div>\n<\/div>\n<div id=\"title_text-block_62178234d05dd\" class=\"w-row title_text\" data-ix=\"slidein\">\n<div class=\"w-col w-col-8 w-col-stack\">\n<article class=\"content-article\">Scientists have found that as we age, the concentration of\u00a0<strong>NAD+<\/strong>\u00a0in cells and tissues gradually\u00a0<strong>declines<\/strong>, a decline that appears to contribute to various\u00a0<strong>diseases<\/strong>\u00a0including cognitive decline, cancers, metabolic diseases, bone fragility and sarcopenia.<\/article>\n<article><\/article>\n<article>\n<div id=\"attachment_905\" style=\"width: 1034px\" class=\"wp-caption aligncenter\"><img fetchpriority=\"high\" fetchpriority=\"high\" decoding=\"async\" aria-describedby=\"caption-attachment-905\" class=\"wp-image-905 size-large\" src=\"https:\/\/solongevity.com\/wp-content\/uploads\/2022\/11\/La-struttura-molecolare-di-NAD-1024x561.jpg\" alt=\"\" width=\"1024\" height=\"561\" srcset=\"https:\/\/solongevity.com\/wp-content\/uploads\/2022\/11\/La-struttura-molecolare-di-NAD-1024x561.jpg 1024w, https:\/\/solongevity.com\/wp-content\/uploads\/2022\/11\/La-struttura-molecolare-di-NAD-300x164.jpg 300w, https:\/\/solongevity.com\/wp-content\/uploads\/2022\/11\/La-struttura-molecolare-di-NAD-768x421.jpg 768w, https:\/\/solongevity.com\/wp-content\/uploads\/2022\/11\/La-struttura-molecolare-di-NAD.jpg 1280w\" sizes=\"(max-width: 1024px) 100vw, 1024px\" \/><p id=\"caption-attachment-905\" class=\"wp-caption-text\">La struttura molecolare di NAD<\/p><\/div>\n<\/article>\n<article class=\"content-article\">Over the past two decades, researchers\u00a0<a href=\"https:\/\/www.nature.com\/articles\/s41580-020-00313-x\">have shown<\/a>, first in animal models and then in some clinical studies, that levels of this molecule<strong>\u00a0can be increased<\/strong>, for example, by appropriate supplementation. The idea is that\u00a0<strong>supplements<\/strong>\u00a0containing the precursors of the NAD+ molecule, i.e., molecules used by the body to produce NAD+ such as the different forms of vitamin B3 (niacin, nicotinamide or NAM and nicotinamide riboside or NR) or nicotinamide mononucleotide (NMN), can help to raise its levels and counteract the effects of various diseases, as well as\u00a0<strong>increase<\/strong>\u00a0the length of life and, in particular, a\u00a0<strong>healthy life<\/strong>.<\/p>\n<blockquote><p>NAD+ influences a number of cell functions, from metabolism to DNA repair, immune\u00a0cell function and\u00a0ageing.<\/p><\/blockquote>\n<\/article>\n<\/div>\n<\/div>\n<div id=\"title_text-block_6217835ed05e0\" class=\"w-row title_text\" data-ix=\"slidein\">\n<div class=\"col w-col w-col-4 w-col-stack\">\n<h3 class=\"article-title\">NAD+ and sirtuins<\/h3>\n<\/div>\n<div class=\"w-col w-col-8 w-col-stack\">\n<article class=\"content-article\">In these early studies, supplement intervention has been shown to be safe and associated with\u00a0<strong>benefits<\/strong>\u00a0against age-related diseases and in slowing aging. High levels of NAD+, in fact, appear to\u00a0<strong>activate<\/strong>\u00a0<a href=\"https:\/\/solongevity.com\/en\/news-en\/sirtuins-the-7-genes-that-make-us-live-longer\/\">sirtuins<\/a>, a group of 7 proteins (produced by the corresponding set of genes called SIRTs) involved in regulating the aging of cells, which use NAD+ itself to carry out their functions. Sirtuins generally become activated under conditions of a metabolic threat and stress (e.g., in the presence of cold temperatures or shortages of nutrients such as amino acids and sugars), reducing cell reproduction activities and thus allowing the organism to focus on\u00a0<strong>survival<\/strong>.<\/p>\n<h3><span style=\"color: #333333; font-size: 30px;\">NAD precursors<\/span><\/h3>\n<\/article>\n<\/div>\n<\/div>\n<div id=\"title_text-block_62178379d05e1\" class=\"w-row title_text\" data-ix=\"slidein\">\n<div class=\"w-col w-col-8 w-col-stack\">\n<article class=\"content-article\"><strong>NAD+ precursors<\/strong>\u00a0are found in numerous<strong>\u00a0foods<\/strong>\u00a0of both\u00a0<strong>plant<\/strong>\u00a0(e.g., spinach, peanuts, avocado, broccoli, cabbage, and watermelon) and\u00a0<strong>animal<\/strong>\u00a0(such as cow\u2019s milk, white meat, salmon, swordfish, and tuna) origin. They are the subject of various\u00a0<strong>studies<\/strong>\u00a0testing their potential as components of<strong>\u00a0dietary supplements<\/strong>\u00a0to restore NAD+ levels, since diet alone may not be enough, and have been shown to be\u00a0<strong>stable<\/strong>\u00a0and\u00a0<strong>safe<\/strong>.Supplementation with these \u201c<strong>NAD boosters<\/strong>\u201d in laboratory research has shown very\u00a0<strong>promising<\/strong>\u00a0results, for example in animal models with obesity, diabetes and cardiovascular disease there have been benefits in terms of\u00a0<strong>weight reduction<\/strong>, increased\u00a0<strong>glucose tolerance<\/strong>\u00a0and improved\u00a0<strong>survival<\/strong>\u00a0rates. There is also evidence that they help\u00a0<strong>counteract neurodegeneration<\/strong>, with increased\u00a0<strong>memory<\/strong>, learning ability,\u00a0<strong>motor function<\/strong>, mitochondrial function, and<strong>\u00a0reduced neuron loss<\/strong>. They also appear to be able to prevent age-dependent changes in\u00a0<strong>gene expression<\/strong>.
